Our facility has added two pieces of equipment that can significantly lower the price of DNA testing, achieve accurate, reproducible results, the BioMek^® 2000 and the ABI Prism^® 3100. Both of these machines reduce labor cost and reagent cost while adding increased accuracy and reliability.

BioMek^® 2000 DNA isolation:
Liquid-handling operations, such as pipetting, diluting, and dispensing, are undergoing major changes in today's life science laboratories in response to increased demands for higher productivity and reduced operator interaction. These basic, everyday functions are being improved significantly by the BioMek systems that performs more efficiently, uses less labor, and leads to increased accuracy and precision.

One of the most time consuming liquid-handling tasks involved with DNA testing is DNA isolation. DNA isolation is very labor intensive and and requires multiple steps. The Biomek offers a degree of accuracy, speed, and reproducibility that is unattainable by traditional manual laboratory methods to isolated DNA. The Biomek liquid handling robot is capable of automatically performing many of the routine procedures required to isolated DNA. Therefore, our scientist do not need to spend their time isolating DNA. Since we do not have to pay the scientist to isolate the DNA by hand we can reduce the cost of the DNA test.

Complete Automation: The 3100 system offers continuous, unattended operation with automated polymer loading, sample injection, separation and detection, and data analysis. After placing plates on the autosampler and importing sample information, the scientist needs only to select the "Start Run" command. The instrument requires no further attention for up to 24 hours.

Higher Sample Throughput: A parallel-capillary configuration gives the ABI Prism 3100 Genetic Analyzer the capacity to simultaneously analyze 16 samples with complete automation. The ABI 3100 can process 16 samples every 30 minutes, therefore, we can analyze about 750 samples in a single day.

Reduced Running and Labor Costs: Automated polymer and sample loading on the 3100 system completely eliminates the time and expense required to pour conventional polyacrylamide and agarose slab gels or to load samples for electrophoresis. Minimal instrument set-up time allows valuable laboratory resources to focus on other critical activities. The increased efficiency and sensitivity of a capillary electrophoresis system reduces the amount of DNA required for each sample injection and this, along with labor savings, reduces the overall cost per sample compared to slab gel systems.

Capillary Electrophoresis: Capillary electrophoresis (CE) is the industry-standard method for DNA fragment separation. The use of CE for DNA analysis offers a number of performance advantages. The efficient heat dissipation of capillaries permits higher run voltages, resulting in faster run times. CE enables the use of electrokinetic injection to load samples into the capillaries. The process of electrokinetic injection involves transfer of negatively charged ions onto the capillary separation matrix via an electromotive force. Because only negatively-charged ions transfer in this process, no significant loss of sample volume occurs. Samples are simultaneously injected into the parallel sixteen capillary array in less than 30 seconds. Using capillary electrophoresis with a sensitive detection system requires very small amount of DNA. The most dramatic advantage of capillary electrophoresis, however, is the elimination of manual operations, resulting in increased run-to-run consistency and reliability.

The AmpFLSTR^® Identifiler^® PCR Amplification Kit

Designed for forensic, paternity testing and research, the Identifiler kit is the only Applied Biosystems’ AmpFLSTR^® product that combines in one PCR amplification the 13 STR loci required by the CODIS database, as well as the Amelogenin gender-determining marker and two additional STR loci. Because the Identifiler kit requires only one PCR reaction, it substantially reduces the workload and corresponding increases the throughput.

The AmpFLSTR^® Identifiler^® PCR Amplification Kit is part of a complete system for DNA fragment analysis that uses five-dye fluorescence technology. Five dyes markedly increase the number of DNA fragments that can be run in a single injection or lane using Applied Biosystems ABI PRISM^® genetic analyzers. This, in turn, substantially increases the number of biological samples that can be processed per day.

This combination of loci meets the recommendation of many international forensic databases. Besides CODIS, the Identifiler kit is also consistent with the standards set by ENFSI (European Network of Forensic Science Institutes), Interpol (International Criminal Police Organization) and GITAD (Grupo Iberoamericano de Trabajo en el Análisis del DNA) for human DNA identification. Intrajurisdictional and even international concordance is now possible, and results can be more easily compared. Besides ensuring concordance with prior forensic tests, sequence consistency also helps maintain a small amplicon size, which is necessary for robust results from challenged or degraded samples.
